Protein Encapsulation in Drug Discovery

Maintenance of drug concentration in the bloodstream or tissue is the crucial factor for drug formulation and drug dosage determination. In general, there are two methods for drug concentration maintenance, which include multiple drug administration at predefined intervals and a single administration of a long-lasting drug formulation. Compared with the former, the latter is a novel approach that reduces the frequency of drug administration. For protein drugs, protein encapsulation in a micro- or nanocarrier is a common approach to achieve a constant release rate over time. After administration, the protein drug can be released from the polymer matrix.

Microfluidic-Based Protein Encapsulation

In general, poly (lactic acid) and poly (glycolic acid) and their copolymers are the most common polymers used for protein encapsulation. The rate of polymer degradation rate depends on the polymer molecular weight and its hydrophobicity. In addition, the particle size is also important for the drug release rate from the particles. In this case, the particles with narrow size distribution are essential to accomplish a reliable and predictable sustained release drug delivery system. As previously described, microfluidic systems present great advantages for the production of the narrow-sized microsphere. In order to encapsulate proteins inside the microspheres, the double emulsion technique and a water-in-oil-in-water (w/o/w) emulsion generated from a microfluidic system are necessary to maximize the encapsulation efficiency.
